The bank was founded in 1945 as Banco Industrial Colombiano (Industrial Bank of Colombia). In 1998 it merged with Banco de Colombia to form Bancolombia. In 2005, Conavi Banco Comercial y de Ahorros S.A. and Corporación Financiera Nacional y Suramericana S.A. were absorbed. In 2007, Banagrícola, a Panamanian bank with a branch in El Salvador, was bought. In 2013, Banistmo and Grupo Agromercantil were bought. Since 1981, the bank’s shares have been listed on the Colombian Stock Exchange, and in 1995 the American depositary receipts were listed on the New York Stock Exchange.

As of 2020, the bank’s network consisted of 1,057 branches, including 666 in Colombia (Bancolombia), 154 in Guatemala (BAM), 92 in Panama and El Salvador (Banco Agrícola), and 6,124 ATMs. Assets at the end of the year were 256 trillion Colombian pesos ($74 billion), of which loans were 191 trillion and investments in securities were 29.6 trillion (mostly government bonds of Colombia and other countries). Deposits taken amounted to 181 trillion pesos.
